28.02.2019: Fully automated dynamic clamp on the Patchliner add on available soon!

After some time of optimisation, we are pleased to announce that the dynamic clamp add-on for the Patchliner which is fully automated and integrated into the Patchliner platform will soon be available! Recent experiments show the reliability of the system, where 8 cells could be recorded simultaneously using dynamic clamp. Could this be the key to recording stable action potentials from hiPSC-CMs?

Using stem cell-derived cardiomyocytes received from our partners Ncardia or Cellular Dynamic International, we could record stable action potentials by introducing electronically modelled IK1 into the cells. Furthermore, full concentration response curves for BayK 8644 were performed on single cells. We saw a concentration-dependent prolongation of the action potential for this compound, as expected for an CaV enhancer. The opposite was true of the CaV blocker, nifedipine, where we saw shortening of the action potential.

This has been a fantastic project with our collaborator Teun de Boer from UMC Utrecht to whom we owe enormous thanks. We're looking forward to bringing this add-on for the Patchliner to our customers for reliable action potential recordings from stem cell-derived cardiomyocytes later this year.
