Background and Assay Description:
We have performed a single solution exchange workflow to activate Clc by 50 mM Chloride concentration jump at pH 5.2 (dark blue trace). By supplementing 100 μM OADS in both measurement solutions (Chloride-free and Chloride containing activating solution), we proceeded to measure the remaining current after inhibition by OADS (red trace). Afterwards, we used the solutions without inhibitor again to wash out OADS from the sensor. The current signal did recover to about 50 % of the original amplitude.
