Background and Assay Description: 

We analyzed the effects of the duration and frequency of the light pulses on the stability of the ChR2 signals. When ChR2 is activated repetitively, the current signals decay faster when time of light and frequency increases. On the other hand, the signal to noise is slightly decreased for time of lights below 10 ms. In terms of signal stability and signal-to-noise, the best assay condition for H134R ChR2 is 2 ms exposure time at 1 Hz. It has to be noted that other ChR2 variants show different optimal conditions, e.g. for T159C ChR2 an exposure time of 20 ms is required to record a full activation signal.
