Background and Assay Description: 

We have performed an assay on ChR2 activity using whole cells on the SURFE2R N1 sensor chips. Cells were detached directly before use and suspended in assay buffer. We used different volumes of cell suspension during sensor preparation and found that 80 μl of cell suspension yield the maximum signal-to-noise during the SURFE2R experiment. With lower volume the cells are more stably adhered to the sensors, but the sensor surface is not fully covered with cells, while at higher volume the cells are not stably attached.
