Title: Functional investigation of GABA and Glutamate re-uptake transporters EAAT3 and GAT1 using SSM-based electrophysiology


GAT1 (SLC6A1) and EAAT3 (SLC1A1) are secondary active neurotransmitter transporters coupled to the ion gradients established by the NaK-ATPase. Both are expressed in neurons and play a major role in the depletion of neurotransmitters GABA and glutamate from the synaptic cleft, thereby taking part in termination of synaptic transmission, maintaining the ambient extracellular neurotransmitter concentration and neurotransmitter recycling through reuptake. GAT1 is linked to epilepsy, schizophrenia, anxiety and ADHD. Subtype specific inhibition of EAAT3 is assumed to be beneficial during phases of insufficient energy supply by preventing reversal glutamate transport. Therefore, screening compounds to find inhibitors of these transporters is of high pharmacological interest. However, there are few methods suitable for resolution of small transporter currents, in particular such providing necessary throughput for compound testing.

Title: Characterization of ion channel currents endogenously expressed in Neuro2A cells using automated patch clamp


Neuro2A cells are a mouse neuroblastoma cell line used extensively to investigate neuronal differentiation, axonal growth and cell signaling pathways. They are also used as an expression system for studying ion channels. Neuro2A cells have been shown to endogenously express NaV channels, predominantly NaV1.7 but also NaV1.2, 1.3 and 1.4 (Lou et al., 2005), mechanosensitive channels Piezo 1 (Coste et al., 2010; Rotordam et al., 2019), purinergic receptors (Vetter & Lewis, 2010) and glutamate receptors (Van der Valk & Vijverberg, 1990).

Title: The role of LRRC8 in the hypotonic stress response of human keratinocytes


The human skin is constantly exposed to various stress factors such as temperature changes, mechanical stress, different humidity levels, air pollution or radiation. These factors can have a tremendous impact on the skin and can contribute to barrier disruption and inflammation, dry and fragile skin as well as premature ageing. Recent advances in different research areas point to an important role of LRRC8 volume regulated anion channels (VRACs) in a plethora of different physiological processes. The function of LRRC8 has been characterized in human keratinocytes and in the native human epidermis and the LRRC8 ion channel has been proposed to be a novel molecular target to modulate keratinocyte differ¬entiation. LRRC8A (also named SWELL1) has been identified as the first essential component of VRACs in various cell types. LRRC8A is composed of four transmembrane domains and a C-terminal domain containing up to 17 leucine-rich repeats. Together with four additional LRRC8 family members (LRRC8B-E) it assembles into hetero-hexameric complexes. The LRRC8 subunit composi¬tion differs between cell types and influences VRAC prop¬erties such as inactivation kinetics, voltage-dependence and selectivity of the transported osmolyte. The generation of LRRC8A-/- knockout HaCaT keratinocytes has provided evidence for the essential function of LRRC8A in hypotonic stress response of human keratinocytes.

About Europhysiology 2022

Europhysiology 2022 marks the continuation of an ongoing and successful partnership between The Physiological Society, The Scandinavian Physiological Society, The German Physiological Society, and the Federation of European Physiological Societies. The series of meetings was initiated with an excellent kick-off in London in 2018. The canceling of the 2020 meeting in Berlin, as a result of the Covid-19 pandemic, has made us even more eager to move Europhysiology 2022 forward with the highest quality program we can muster.

We look forward to welcoming scientists at all stages of their careers; from students to postdocs, principal investigators, and established leaders within Physiology who will inspire us through invited lecturers. Importantly, the first day of the conference (September 15th) will feature pre-meetings of “Special Interest Groups” within more specific topics, providing even more scientific depth. Thus, we promise a unique scientific experience, unparalleled in our field.
