According to the transport direction of 2Cl-/1H+ exchange, we observed positive (Cl- efflux) or negative (Cl- influx) transient currents.
Background and Assay Description:
We have analyzed transporter symmetry by comparing chloride influx with chloride efflux. In both cases, we prepared proteoliposomes in Cl-free solution. For the influx assay, we activated Clc by a single solution exchange providing 20 mM chloride. This leads to a negative current, reflecting the chloride induced net movement of one negative charge into the vesicles. For the efflux assay, we have equilibrated the sensor in solution containing 20 mM chloride, followed by Clc activation by removing chloride.
