Session Description:

This joint partner session will focus on the characterization of drug effects on human induced pluripotent stem cell-derived (hiPSC-derived) cardiomyocytes and hepatocytes. The use of these cell models continues to increase in the assessment of cardiac safety and toxicological side effects of newly developed compounds, due to their reproducibility and low ethical concern.

However, the obstacles derived from using a 2D culture setting, immature phenotype, or serum-containing media for chronic assays raise concerns over non-physiological responses in preclinical drug development and safety. Here, we will introduce solutions to these hurdles by using innovative pro-maturation technologies, specialized cell culture conditions and newly developed recording protocols. Characterization, pro-maturation and toxicity assays using iCell Cardiomyocytes2 for automated patch clamp (Patchliner Dynamite8) and contractile force recordings (FLEXcyte 96) will be presented.

Furthermore, data highlighting acute and chronic effects of metabolically active hepatotoxic compounds (Aflatoxin B1) will be shown in standard 2D conditions using an impedance-based, non-invasive assay (CardioExcyte 96), indicating that the iCell Hepatocytes are metabolically active and functional.
